Do you have a minute?

Minute的意思是：分钟；片刻，所以“A minute”在不同的语境下既可以理解为“一分钟”，也可以理解为“（笼统的）一段时间”。所以想问别人“你有空吗”，就可以说：Do you have a minute? 另外，助动词Do也可以省略，直接说成You have a minute?
例句：
You have a minute? I want to discuss this issue with you. 
你现在有空吗？我想和你讨论一下这个问题。


Are you available?

Available的意思是：可会见的、可与之交谈的；由此可以衍伸为“有空的”的意思；没空就是“Not available”
例句：
Are you available? I need your help.
你有空吗？我需要你的帮忙。
Sure. 
当然可以。


Can you spare some time?

Spare作动词时，有“匀出，抽出”的意思，所以可以使用Can you spare some time来问别人“你有时间吗”
例句：
Can you spare some time to go shopping with me?

When do you have time off?

Have time off是个固定搭配，意思是“休息，有空，放假”，所以When do you have time off？就是问别人“你什么时候有空？”，是不是很容易记住呢？

When will you get around to something?

这是个固定的表达方式，Get around to (doing) sth的意思是“抽时间做某事”。
例句：
When will you get around to traveling? 
你什么时候有空去旅游？


When do you find time to do something?

与Get around和Spare time一样，Find time也是“抽出时间、匀出时间”的意思，属于同义表达。
例句：
When do you find time to talk to her? She is waiting for your reply. 
你什么时候有时间和她谈谈？她一直在等你的答复。
